Grundlage fUr dieses Buch sind meine Vorlesungen in Immunbio logie, die ich in den vergangenen Jahren an der Purdue Universitiit gehalten habe. Ziel der Vorlesungen ist ein aktueller Oberblick iiber die Biologie der Immunantwort und nicht eine vollstiindige Zusammenfassung aller Aspekte der Immunologie. Schwerpunkte dieses Buches sind Wechselwirkungen zwischen Zellen und Fragen der Regulierung der Immunantwort, vor allem die Rolle des MHC bei den Wechselwirkungen zwischen Zellen, die Bedeutung der Autoreaktivitiit gegen MHC-Antigene, die Regulierung der Im munantwort durch ein Netzwerk von anti-ldiotypen und Regel kreise bei der Suppression. Dariiber hinaus gehe ich auf die faszi nierenden neuen molekularbiologischen Erkenntnisse iiber die Immunglobulin-Gene und die Entstehung der Antikorper-Vielfalt ein. Das Hauptthema ist die Voraussetzung von Selbst-Erkennung und Selbst-Reaktivitiit fiir die Entstehung von Immunreaktionen. 1m Verlauf der Vorlesung versuche ich, moglichst viele Probleme anhand von Versuchsprotokollen zu erliiutem und, wenn moglich, die geschichtliche Entwicklung der Experimente zu beschreiben. Auch in diesem Buch habe ich versucht wiederzugeben, wie immu nologische Fragen wissenschaftlich angegangen werden. Ich habe mich bemiiht, moglichst keine Langeweile aufkommen zu lassen. Sicherlich werde ich viele Freunde und Kollegen iirgem oder schockieren durch die Art und Weise, wie ich durch die Auswahl bestimmter Experimente die Punkte zu verdeutlichen versuche, auf die es mir ankommt. Ich mochte mich gleich hier zu Beginn des Buches bei ihnen entschuldigen und kann nur hoffen, daB sie am Ende des Buches wieder versohnt sind."
