La caract�risation structurelle du domaine kinase ALK et les informations sur les interactions m�dicament-prot�ine pourraient �tre tr�s utiles dans la conception rationnelle de nouveaux m�dicaments sp�cifiques pour le traitement de l'ALCL ALK+ et d'autres tumeurs ALK+. Ce travail s'est concentr� sur la production et la purification de diff�rentes formes du domaine kinase ALK afin d'obtenir de bons candidats pour des �tudes structurales ALK par cristallographie aux rayons X et STD-NMR. La meilleure forme de prot�ine recombinante a �t� utilis�e pour cribler les inhibiteurs ALK potentiels et pour d�montrer l'interaction entre le domaine kinase ALK et un nouvel inhibiteur de kinase. Les r�sultats de ces travaux pourraient ouvrir la voie au d�veloppement de nouveaux m�dicaments sp�cifiques pour le traitement des maladies ALK+. Ces travaux pourraient fournir un mod�le m�thodologique pour la production et la purification de prot�ines recombinantes et pour les prochaines �tudes sur les interactions entre m�dicaments et prot�ines.
