Ces dernières années, grâce aux progrès réalisés dans l'humanisation des anticorps monoclonaux recombinants, ceux-ci ont vu leur utilisation thérapeutique s'accroître. Le rituximab a montré son efficacité dans le traitement des lymphomes de faible et de haute malignité. Néanmoins 30 à 50% des patients ne répondent pas au traitement. Dans une première partie nous avons défini les conditions expérimentales permettant de reproduire chez la souris le développement d'un lymphome agressif et disséminé. Dans une seconde partie nous avons étudié l'effet thérapeutique du rituximab sur ce lymphome. Nous avons retrouvé la grande variabilité de réponse au rituximab. Nous avons montré qu'il existait une relation entre le volume tumoral et la réponse au traitement. Nous avons pu construire un modèle concentration/effet nous permettant de démontrer l'existence d'une relation entre l'efficacité du rituximab et le volume tumoral. Enfin dans un troisième volet nous avons montré l'important effet rayonnant de particules d'oxydes de gadolinium et de bore lors d'une irradiation sous un faisceau de neutrons après une étape d'internalisation des particules au sein des cellules.
