Les Sphingolipides sont des lipides complexes pour lesquels une dérégulation du métabolisme a été décrite dans de nombreuses pathologies, telles que le syndrome métabolique et de nombreuses maladies génétiques. Dans le foie, les enzymes impliquées dans le métabolisme des lipides sont très sensibles aux régulations transcriptionnelles. Du fait que les acides gras soient nécessaires à la synthèse des sphingolipides, nous nous sommes demandés si les récepteurs nucléaires impliqués dans la régulation transcriptionnelle de leur métabolisme l'étaient également dans celle des sphingolipides. Dans cette étude, nous avons testé l'hypothèse que les récepteurs nucléaires (LXR et PPAR), principaux régulateurs transcriptionnels de la biosynthèse des acides gras hépatiques et de la dégradation, peuvent influencer le métabolisme des sphingolipides. Pour répondre à cette hypothèse, nous avons utilisé une approche pharmacologique in vivo pour induire l'activité de LXR et PPAR chez des souris de type sauvage et des souris transgéniques déficientes en LXR ou en PPAR. Pour la première fois, nos données montrent que le métabolisme des sphingolipides semble ètre fortement touché par LXR et PPAR.
